Angels are supposedly emotionless, which means they shouldn't be able to ~fall in love~ with each other.
(warning: i'm rambling and this will probs not make sense)
ok i'm kinda ambivalent about that reasoning because imo angels are certainly capable of emotion. now, this doesn't mean an angel's emotions are like a human's. so they might not even experience romantic love. i mean, cas became clearly attached to dean even before he fell, but i'm sure that romantic love had anything to do with that. so he maybe came to love him like he loves the rest of his brothers and sisters. idk.
but then cas falls (or something like it) and you could make a case for him developing human emotions now that he's cut off from heaven's influence (and i think misha has been pretty clear that that's exactly what's happening).
and then you have anna. i definitely noticed that she became more serious/businesslike/cold/whatever after regaining her grace, but i'm not sure she forgot about how it felt to have human emotions (and a female human body). the writers could take the "she still remembers her humanity" route to make anna/cas happen. more so if she falls out of favor with heaven again and ends up sort-of falling too.
i don't know which option would cause me to die of disgust faster: if they paired them up and completely glossed over the incest factor or came up with some bullshit "oh they're an exception and this is why" hand-wave. because it WILL smell like bullshit, no matter what they say to make it not-creepy.
and there's also what i told ibroketuesday: the platonic brother/sister dynamic or even the commandant/subordinate soldier dynamic is much more interesting to explore than just another boring hetero romance blah blah etc.
i like anna (or, rather, i like the *idea* of anna) but if her coming back means cas/anna, i'm going to have to say no thx. i'd roll my eyes if it means more dean/anna too (i'd prefer she comes back to kick ass instead of making cow eyes at dean, even if i find the idea of dean/anna cute in theory) because yeah, plz to be giving women a role that does not include "love interest", but it wouldn't make me see red like cas/anna would.
